- 目的 观察不同灸法对环磷酰胺所致免疫抑制兔脾组织程序性死亡因子-1(PD-1)及其配体PD-L1表达的影响。方法 大耳白兔32只随机分为空白组、模型组、隔药饼灸组、艾灸组。除空白组外,其余3组腹腔注射环磷酰胺制备免疫抑制兔模型,连续7 d。成模后,隔药饼灸组和艾灸组分别给予隔药饼灸、艾灸干预,隔日灸,共灸10次。治疗结束后次日动物麻醉,摘取脾脏。采用RT-qPCR检测PD-1、PD-L1 mRNA表达,免疫组化检测PD-1蛋白表达。结果 与空白组比较,模型组兔脾组织PD-1、PD-L1 mRNA表达明显升高(P<0.01);与模型组比较,隔药饼灸组兔脾组织PD-1、PD-L1 mRNA表达明显降低(P<0.01),艾灸组兔脾组织PD-L1 mRNA表达明显降低(P<0.05),PD-1 mRNA表达虽有降低趋势但差异无统计学意义(P>0.05)。免疫组化结果显示,兔脾组织棕褐色PD-1阳性表达主要在细胞浆、细胞膜或间质,与空白组比较,模型组兔脾组织PD-1蛋白表达显著升高(P<0.01);与模型组比较,隔药饼灸组和艾灸组兔脾组织PD-1蛋白表达显著降低(P<0.01,P<0.05)。结论 不同灸法对免疫抑制兔免疫功能均有明显改善作用,隔药饼灸较艾灸更具优势。
- Abstract:
- Objective To observe the changes of expressions of PD-1 and PD-L1 in immunosuppressive rabbits induced by cyclophosphamide through different types of moxibustion. Methods Totally thirty-two rabbits were randomly divided into blank group , model group , herbal cake-separated moxibustion group and moxibustion group. Except the blank group, the other three groups were intraperitoneally injected with cyclophosphamide to make immunosuppressive rabbit model for 7 days. After the model was established , herbal cake-separated moxibustion group and moxibustion group were treated with herbal cake-separated moxibustion and directed-moxibustion respectively , once every other day , 10 times in total. The next day after the treatment , the animals were anesthetized and the spleen was removed. The expressions of PD-1 and PD-L1 mRNA were detected by real-time fluorescence quantitative PCR , and the expression of PD-1 protein was detected by immunohistochemistry. Results Compared with the blank group, the expressions of PD-1 and PD-L1 mRNA in the spleen tissue of the model group increased significantly (P<0.01). Compared with the model group, the expressions of PD-1 and PD-L1 mRNA in the spleen tissue of the rabbits in the herbal cake-separated moxibustion group was significantly lower (P<0.01), and the expression of PD-L1 mRNA in the spleen tissue of the moxibustion group was significantly lower (P<0.05), while the PD-1 mRNA showed a decreasing trend , without statistical significance (P>0.05). Immunohistochemical results showed that the positive expression of PD-1 in rabbit spleen tissue was mainly in the cytoplasm, cell membrane or interstitial. Compared with the blank group , the expression of PD-1 protein in the rabbit spleen tiss ue of the model group significantly increased (P<0.01). Compared with the model group, the expression of PD-1 protein in the spleen tissues of rabbits in the herbal cake-separated moxibustion group and moxibustion group was significantly reduced (P<0.01, P<0.05). Conclusion Different types of moxibustion have obviously improved the immune function of immunosuppressive rabbits , and herbal cake-separated moxibustion has more advantages than moxa moxibustion.
