补肾方药干预皮质酮所致早发性卵巢功能不全大鼠比较研究

- 目的 比较补肾活血方、左归丸和右归丸3种补肾方药干预皮质酮所致早发性卵巢功能不全效果。方法 50 只雌性SD大鼠随机分为正常组、模型组、补肾活血方组、左归丸组、右归丸组,每组10只。皮下注射皮质酮建立早发性卵巢功能不全大鼠模型。各给药组给予相应药物干预,3周后取大鼠卵巢组织,检测血清雌二醇、卵泡刺激素、抗苗勒管激素浓度,评价卵巢功能。左侧卵巢组织HE染色观察卵巢形态,镜下进行卵泡计数;Western blot检测右侧卵巢组织Angptl4和Np4蛋白表达。结果 与正常组比较,模型组大鼠卵巢组织血清雌二醇、抗苗勒管激素浓度降低,卵泡刺激素浓度升高;卵巢颗粒细胞排列紊乱,闭锁卵泡增多,其他各级卵泡减少,Angptl4和Np4蛋白表达升高。经补肾方药干预后上述情况均有不同程度改善,其中补肾活血方效果最优。结论 3 种补肾方药均能治疗皮质酮所致早发性卵巢功能不全,其机制可能与下调卵巢组织Angptl4和Np4的表达相关。
- Abstract:
- Objective To compare the effects of Bushen Huoxue Prescription, Zuogui Pills and Yougui Pills on the treatment of corticosterone induced premature ovarian insufficiency. Methods Totally fifty female SD rats were randomly divided into normal group, model group, Bushen Huoxue Prescription group, Zuogui Pills group and Yougui Pills group, with 10 rats in each group. Premature ovarian insufficiency rat model was established by subcutaneous injection of corticosterone. Each administration group was given relevant medicine for intervention. Three weeks after modeling and administration, ovarian histological analysis and the concentrations of estradiol, follicle stimulating hormone, and anti-Mullerian hormone were used to evaluate ovarian function. The ovarian morphology and count follicles with HE staining microscopy on the left ovary tissue were observed. Western blot was used to detect the expression of Angptl4 and Np4 protein in the right ovarian tissue. Results Compared with the normal group, the concentration of estradiol and anti-Mullerian hormone decreased in the model group, and the follicle stimulating hormone concentration increased; the ovarian granulosa cells were disorderly arranged, the atretic follicles increased, the follicles at other levels decreased, and the expression of Angptl4 and Np4 protein increased. The above conditions were improved after the intervention of the three tonifying kidney prescriptions, and the effect of Bushen Huoxue Prescription was the best. Conclusion The three prescriptions can treat corticosterone induced premature ovarian insufficiency. The mechanism may be related to the down-regulation of expressions of Angptl4 and Np4 in ovarian tissue.
