- 目的 观察电针对SAMP8小鼠皮质及海马突触素(SYN)和突触后致密物-95(PSD-95)表达的影响,探讨电针治疗阿尔茨海默病的介入时机。方法 SAMP8 小鼠48只随机分为模型组、3月龄电针组、6月龄电针组和9月龄电针组,每组12只,选取同龄正常老化SAMR1小鼠12只为对照组。电针组电针“百会”“大椎”“肾俞”,频率2 Hz,每日1次,8 d为1个疗程,疗程间隔2 d,共3个疗程。各组均在10月龄取材,选择皮质及海马区,采用免疫组化和Western blot检测突触相关蛋白SYN、PSD-95的表达。结果 与对照组比较,模型组小鼠皮质及海马SYN和PSD-95蛋白表达明显降低(P<0.01);与模型组比较,各电针组小鼠皮质及海马SYN、PSD-95蛋白表达明显升高(P<0.05,P<0.01);与3月龄电针组比较,6月龄电针组、9月龄电针组皮质及海马SYN、PSD-95蛋白表达明显降低(P<0.05,P<0.01)。结论 电针可促进SAMP8小鼠皮质及海马SYN、PSD-95蛋白的表达,改善突触功能,且早期电针治疗效果最明显。
- Abstract:
- Objective To observe the effects of electroacupuncture on the expressions of synaptophysin (SYN) and postsynaptic density-95 (PSD-95) in the cortex and hippocampus of SAMP8 mice at different stages; To explore the timing of electroacupuncture in the treatment of Alzheimer disease. Methods Totally 48 SAMP8 mice were randomly divided into model group, 3-month-old electroacupuncture group, 6-month-old electroacupuncture group and 9-month-old electroacupuncture group, with 12 mice in each group. 12 SAMR1 mice of the same age of normal aging were set as control group. “Baihui”(GV20), “Dazhui”(GV14) and “Shenshu”(BL23) acupoints were selected in the electroacupuncture groups. The frequency of electroacupuncture was 2 Hz, once a day, and 8 days was a course of treatment. Samples were collected from each group at the age of 10 months, and the protein expressions of SYN and PSD-95 in cortex and hippocampus were detected by immunohistochemistry and Western blot. Results Compared with the control group, expressions of SYN and PSD-95 protein in the cortex and hippocampus of the model group decreased significantly (P<0.01). Compared with the model group, expressions of SYN and PSD-95 protein in the cortex and hippocampus of mice in each electroacupuncture group significantly increased (P<0.05, P<0.01). Compared with 3-month-old electroacupuncture group, the expressions of SYN and PSD-95 protein in the cortex and hippocampus of 6-month-old and 9-month-old electroacupuncture groups decreased significantly (P<0.05, P<0.01). Conclusion Electroacupuncture can promote the expressions of SYN and PSD-95 protein incortex and hippocampus of SAMP8 mice, and improve the synaptic function, and early electroacupuncture has the most obvious efficacy.
